はじめに
Introduction

従来の透過吸収分光法では、微量な分子を観測するために、バックグラウンド光の中のわずかな吸収を捉える必要がありました。このとき、レーザーパワーを大きくすることで感度を向上させることができますが、検出器の観測可能な最大値により制限されてしまいます。

ここで着目してほしいのが、検出器が受け取る信号のほとんどは見たい分子の吸収とは関係のないバックグラウンド光であることです。もしこのバックグラウンド光を除去して、見たい分子の吸収のみを観測することができれば、検出器に制限されずに、レーザーパワーの増加にともなって感度を大きくすることができます。

これを実現するのが、私たちが開発しているバックグラウンドフリー分光法です(図1)。

In conventional transmission absorption spectroscopy, it is necessary to capture a small amount of absorption in the background light in order to observe small amounts of molecules. Although the sensitivity can be increased by increasing the laser power, it is limited by the maximum value that can be observed by the detector.

It is important to note that most of the signal received by the detector is background light, which has nothing to do with the absorption of the molecule we want to see. If we can remove this background light and observe only the absorption of the molecule we wish to study, we can increase sensitivity with increasing laser power without being limited by the detector.

This is achieved by background-free spectroscopy, which is what we are developing (Fig. 1).

方法
Methods

では、どうすればバックグラウンド光を消せるのでしょうか。

わたしたちは、光が波の性質を持つことを利用しています。

海岸の波のように、2つの光を重ね合わせると干渉し、強め合ったり弱めあったりします。私たちは、その中でも弱めあいの干渉を利用しています。弱めあいの干渉では、打ち消し合うことで光が出ない状況になりますが、片方の光のみに見たい分子を吸収させると、弱めあいの干渉条件が崩れ、吸収分の光が現れます。

これにより、バックグラウンド光を除去して分子の吸収だけを観測できます。

So how can we turn off background light?

We take advantage of the wave nature of light.

Like waves on a beach, when two lights are superimposed on each other, they interfere, either strengthening or weakening each other. We make use of mutual weakening interference. However, if only one light absorbs the molecule of interest, the weakening interference condition is broken and the light of the absorbed molecule appears.

This allows us to remove the background light and observe only the absorption of the molecule.

結果1:バックグラウンドフリー振動分光法の実証

私たちは、干渉計とよばれる装置を使ってバックグラウンドフリー分光法を実現し、図2のように、分子の吸収信号のみを観測することに成功しました。

We have realized background-free spectroscopy using a device called an interferometer and succeeded in observing only molecular absorption signals as shown in Fig 2.

結果2:マルチプレックス計測による高感度化

バックグラウンドフリー分光法では、分子特有の複数波長で光が発生します。指向性の高いレーザー光源を用いることで信号を大きくしていますが、対象分子が少なくなると、信号が検出器の雑音に埋もれてしまう課題がありました。そこで私たちは、分子の吸収パターンに一致するスペクトルフィルターを用い、複数波長の信号を一括取得する手法(マルチプレックス計測法)を考案しました。これにより、雑音の影響を抑え、分光感度を劇的に向上できることを示しました。

Background-free spectroscopy generates light at multiple wavelengths unique to each molecule. While bright lasers can boost the signal, detecting extremely small amounts of molecules remains difficult because the signal is easily hidden by detector noise. To solve this, we developed a new technique that captures multiple wavelengths at once by utilizing a spectral filter matched to the molecule’s absorption profile. As a result, we demonstrated a dramatic improvement in sensitivity by significantly reducing the effects of noise.
